Expansion
of cell number would be the most important thing for the success of human cell
therapy. During this cell expansion, trypsin dissolution is still necessary to
move cells to new attachment surfaces. However, this trypsin is suspicious as a
strong irritating chemical and mutagen to the cells, and it is still one of the
biggest variable to decide the possibility of genetic transforms. More over the
toxicity, repeated washing and centrifugation, reattachment would make apoxic
period for 20 minutes. These conventional trypsin cultures could not be
physiologic situations. We
used mechanical scraping in water proof vessels having multiple rubber ports would
be good for trypsinless cell expansion with easy monitoring because we can not
only see colony patterns but also fit medical protocols to prevent
contaminations. We expanded the ASCs number 32 times within 3 weeks without
trypsin treatment. For the movement of cells to new surfaces, we just used scraping
with increasing the vessel numbers. When we need the cells for therapy, we
scraped cells mechanically and use directly to patients. As results, we completely
removed the trypsin over 200 times expansion of cell numbers, also we reduced
whole manual processing hours under 20%. ![]() ![]() |
